Think of this as a post. They are and must be 140 characters or less.
This is a way to

reply to someone's tweet
This is like forwarding a tweet. Use a capital RT with a mention to give the original tweeter credit (RT @friend).
Think of this as a private tweet between you and the person receiving the message. You use a capital D with the mention (D @friend).
Adding the # before a word makes that word searchable on twitter.
You can organize your tweets this way or use them to partcipate in a larger conversation.
This is what is popular on twitter right now. A trend can last an hour or a week.
Lists are a way to organize the people that you follow into categories. Lists can be public or private. YOu can also follow other people's lists.
