RAF

  • Rapidly Accelerated Fibrosarcoma
  • Fibrosarcoma: A tumor that derives from fibrous connective tissue.

A-RAF

  • Involved in the transduction of mitogenic signals from cell membrane to nucleus.
  • Serves as a regulator for myogenic differentation by inducing cell cycle arrest, the expression of muscle-specific proteins, and myotube formation.
  • Mutations found in 60 different cancer lines
  • However, may not cause cancer.

BRAF

c-Raf

  • Involved in sending signals inside cells, which help in cell growth
  • 2002, shown to be mutated in human cancers.
  • Inherited BRAF mutations can cause birth defects.
  • Mutations can also appear later in life and cause cancer as an oncogene.
  • AKA: RAF1
  • Similar to BRAF, it helps with cell growth by using cell signals
  • Mutation can cause many different diseases.
  • Can cause LEOPARD Syndrome. (Retarded Growth, Abnormal Genitalia, Deafness, etc..)

MAPK Cascade

RAF Kinase

  • RAF kinases participate in the MAPK pathway.
  • MAPK pathway is a chain of proteins in a cell that communicates a signal from a receptor on the surface of the cell to the DNA in the nucleus of the cell.
  • Mitogen-Activated Protein Kinases.

RAF Family

  • ARAF
  • BRAF
  • c-RAF
