Leaf Disk Assay - Photosynthesis Lab

Bailey Russell, 6

Photosynthetic Effective Time

The rate at which photosynthesis is occurs can be calculated by using 1/PSET-50, which is the amount of time it takes for half of the leaflets to float in the given amount of time. The larger the PSET-50, the slower the rate of photosynthesis; the smaller the PSET-50, the faster the rate of photosynthesis.

6CO2+6H20 - >C6H1206+6O2

Sunlight strikes the chloroplast which excites the molecules so that it loses an electron. A molecule of water is split apart to replace the lost electron. When the water is split, oxygen is produced as a waste product. Six molecules of carbon dioxide are fixed with six molecules of RuBP to produce twelve molecules of PGA. The twelve molecules of PGA are then rearranged into G3P, a sugar (C6H12O6).

Using a Leaf Disk Assay

Photosynthetic rates can be determined over 20 minutes using a leaf disk assay. After removing the air from a syringe containing the leaflets and water, the leaflets will sink, indicating that they are now deprived of air. Pour the leaflets into a sodium bicarbonate container and record for every two minutes the amount leaflets that have floated to the top of the solution. This will help indicate the photosynthetic rate of the leaflets.

Photosynthesis

Plants photosynthesize order to produce sugar and oxygen, which, in turn, gives the plant energy. Photosynthesis usually tends to occur in the leaves where many chlorophyll are located. When the leaves are exposed to light, the process of photosynthesis begins. The more light the plant is exposed to, the quicker the rate of photosynthesis will occur. Altering the amount of carbon dioxide given to the plant can also alter the rate at which photosynthesis occurs.
