Streaking Method
What is streaking?
is a procedure which the inoculum is implanted in a straight line or zigzag line. Streaking process is delicate so doing it gently and lightly is recommended.
How to do it

INOCULUM
- the sample or specimen to be inoculated
In this kind of streaking method, the test identifies bacteria in urine. It is used to diagnose and manage urinary tract infections.

Urine Culture
You need:
Apply loopful amount of bacteria using sterile
inoculating loop.
On UPPER portion of the plated media, start streaking gently and lightly. Lines should not be too close to each other.

- calibration for urine is 1:100 and 1: 1000 microliter
Streak- Plate
- used in anti-microbial susceptibility tests
-also called MULTIPLE-INTERRUPTED
and is used for semi-quantitative determinations.
- there are four divisions that can be seen
2. Rotate, then sterilize the loop. Start at the last point of the first streak.
3. Continue the same routine until quadrant 3. On the 4th, do simple streaking. Usually the 1st and 2nd have more organisms.

HOW?
1. Firstly, get a loopful of organism and streak on the upper portion only. Streak lines should be close to each other. You now have the first quadrant.

To do: Streak using sterile cotton swab all over the plate. Rotate then streak again overlapping the first. Lastly, repeat for the third time.

Simple Streaking
Before starting the process, ALWAYS do the aseptic
technique
BROTH
- in this liquid media, we use the tapping method.
- put a loopful of organism in the broth using an inoculating loop. After incubation turbidity is
observed.
SLANT
- inoculating loop is used here
and simple streaking method is applied.

Tube Media
There are two METHODS used: STAB and STREAK
-the media is butt and slant and stabbing first then simple streaking is applied.
-inoculating needle is used.
