The simplest form of endocytosis is observable among the simplest organisms. One example is a free-living ciliate protozoa, the Tetrahymena sp.

The species of Tetrahymena is widely used in experimental biology due to its microtubular array structure and dual nucleation. Apart from that, several vacuoles are also present in the organism making it a viable tool for mode of cell absorption.

Pinocytosis is also a form of endocytosis. This invagination is different from phagocytosis primarily because the mechanism is the intake of minute particles. The mechanism is called “cell drinking” because the substance to be taken up by the cell will be dissolved in water, the “drinking” term indicates that the substances dissolved in water will be taken up by the cell.

In this experiment, Phagocytosis in Tetrahymena sp, has been observed using two methods. The first involves the introduction of 1% india ink to the protozoan, Tetrahymena sp. In this particular method, pinocytosis was demonstrated. This could be seen through the internal formation of vacuoles filled with ink in the cell. The cell was then exposed to 3% Formalin solution for further examination and to undergo a count of the average number of vacuoles produced by the organism.

The second method used to demonstrate phagocytosis is the introduction of lead shavings or graphite into the cell suspension. Graphite is a mineral that cannot easily be metabolized by the Tetrahymena, which will allow it to be present in the cell for a long period. This method allows one to be able to observe phagocytosis more clearly by allowing the organisms to engulf the solid dark particles.

Phagocytosis is a form of endocytosis in which an organism engulf or ingest a large substance or molecule present in its environment that forms a membrane inside the phagocyte (engulfing cell) with the aid of the plasma membrane.

In the experiment, we identified the pinocytotic mechanism through the action of the organism to India ink, and the phagocytosis through the ingestion of the graphite shavings from the lead of the pencil. It was observed too that the presence of vacuoles or vacuole formation is directly proportional to the concentration and presence of India ink in the medium. The, organism based on the food particle absorbed, prefer the India ink as food source.
