En 1983 utilizó una técnica que consistía

de ADN al medio de reacción.

La longitud de los primers y la secuencia designan los parámetros de una amplificación, una fórmula simple para calcular la Tm (“melting temperature”) es la siguiente:

Las cantidades óptimas de Taq polimerasa necesarias
